Column scale-up

During many instances, it may be necessary to increase the size of a chromatography column in order to increase the production capacity of a process. By doing so will require a scale-up of the column to a suitable size for the job.

The most straightforward way to scale up a column is to maintain the same column length and to increase the cross-sectional area to maintain a constant ratio of unknown to column volume. A simple way to calculate the scaled diameter is given by the following equation:

To recreate identical operating conditions on the scaled column as the unscaled one, the linear flow rate must be kept constant. The volumetric flow rate required to keep a constant linear flow rate can be found by:
